Her first research projects concentrated on mycology, lichenology and plant physiology. Her interests also included genetics applied to ornamental plants, phytopathology and floriculture. She also researched the systems of both spontaneous and cultivated species of plants and flowers.
During her experience as Director of the Botanical Garden in Cagliari, Mameli-Calvino confirmed her pragmatic attitude to work by introducing a series of species that had economic and social value. Noteworthy examples include the Pennisetum purpureum Schum. et Thom., Chloris gayana Kunth, Manihot palmata Müll.Arg. var. aipi (Pohl.) Müll.Arg., Solanum muricatum Ait., Casimiroa aedulis La Llave, etc. At the talk (Index Seminum) she delivered on 22 December 1928 in front of the Academic Senate, Mameli-Calvino remarked that The acclimatization experiments carried out on these plants at the Botanical Garden have yielded excellent results. In particular, while talking about the Elephant grass ((Pennisetum purpureum), Rhodes grass (Chloris gayana) and Indian fig opuntia (Opuntia ficus indica Mill. var. inermis Moris et De Not.), she pointed out that they would solve the problem of lack of forage during the summer month in Sardinia and any other drought areas in Italy.
